Which biomarkers do you know?

All fields are optional. The more detail, the more precise the matching. If you do not know a value, leave "I do not know" — we will show trials without that restriction.

⭐ Critical biomarkers Decisive for matching

MSI ? MSI / MMR MSI-H/dMMR responds very well to immunotherapy (pembrolizumab, nivolumab). MSS is the majority of CRC. Your report shows it after IHC or PCR/NGS.

KRAS ? KRAS The most relevant mutation in CRC. G12C has specific drugs (sotorasib, adagrasib). G13D and others fit pan-RAS like daraxonrasib. The exact variant changes the trials.

NRAS ? NRAS KRAS partner — they form the "RAS pathway". If one is mutated, anti-EGFR (cetuximab, panitumumab) does not work. Your NGS report almost always includes it.

BRAF ? BRAF Subset of 7-10% of CRC. V600E (class I) has approved encorafenib + cetuximab. Classes II/III respond differently. If absent in modern NGS, almost certainly Wild-type.

HER2 ? HER2 / ERBB2 Distinguishing levels is decisive. Positive (IHC 3+ or IHC 2+ FISH+) has T-DXd and trastuzumab+tucatinib. Low (IHC 2+ FISH-) is under research. Negative is most cases.

🧬 Other relevant biomarkers Optional — open more trials if you know them

TP53 ? TP53 The most common tumor suppressor in CRC (~60%). Some trials require it or stratify by it.

PIK3CA ? PIK3CA PI3K/AKT/mTOR pathway. Some mutations allow copanlisib or alpelisib in studies. ~15-20% of CRC.

CCNE1 ? CCNE1 Cyclin E1. Its amplification opens trials with WEE1 inhibitors (adavosertib) and others. Rare but actionable.

TMB

? TMB — Tumor Mutational Burden Tumor Mutational Burden. TMB-H (≥10 mut/Mb) has tumor-agnostic approval for pembrolizumab. Your NGS report usually includes it.

PD-L1

? PD-L1 Some trials require CPS≥1 or ≥10. For CRC less common than for other tumors, but relevant in MSI-H.

🔬 Rare biomarkers Less than 5% of CRC but decisive when present

NTRK ? NTRK Larotrectinib and entrectinib are approved tumor-agnostic for NTRK fusions. Only ~0.5% of CRC but spectacular responses.

RET ? RET Selpercatinib and pralsetinib approved for RET fusion. Very rare in CRC but searchable.

MET ? MET Inhibitors: tepotinib, capmatinib. Amplification or exon 14 skipping. Rare de novo in CRC, more common after anti-EGFR.

FGFR2 ? FGFR2 Pemigatinib, futibatinib. Fusions and amplifications in some basket trials.

POLE ? POLE Hypermutated tumors. Mutations in exonuclease domain generate very high TMB and excellent response to immunotherapy, even in MSS.

BRCA1/2 ? BRCA1/2 PARP inhibitors like olaparib in trials for tumors with HRD. Rare in CRC but relevant in hereditary colorectal.
