Nuestro objetivo fue caracterizar de forma exhaustiva el panorama genómico y las características clínicas de los CCR con mutaciones en ERBB2/HER2 y compararlos con los CCR con amplificación de ERBB2/HER2. Analizamos los datos de cBioPortal de 4.348 pacientes con CCR con perfiles de ERBB2/HER2 disponibles. Se evaluaron las características clinicodemográficas, la inestabilidad de microsatélites (MSI), la carga mutacional tumoral y las alteraciones en 94 genes asociados al cáncer mediante la prueba de chi-cuadrado/Fisher, la prueba de Kruskal-Wallis y la agrupación jerárquica.

Se identificaron mutaciones putativas de ERBB2/HER2 como genes impulsores en 95 pacientes (2,2%), amplificaciones en 87 (2,0%) y estado de tipo salvaje en 4.166 (95,8%). En la cohorte general, los tumores con mutaciones en ERBB2/HER2 demostraron un enriquecimiento significativo de KRAS (50,5%) en comparación con los de tipo salvaje (37,7%; p = 0,014) y los amplificados (18,4%; p < 0,001), junto con un enriquecimiento de MSI (37,2% frente a 1,6%; p < 0,001), una mayor carga mutacional tumoral y una predominancia en el lado derecho.
